Documentation:Standard Operating Procedures for Bio-Rad CFX96 Real-Time System with C1000 Touch Thermal Cycler

1.   Please obtain proper training from the FNH teaching lab technician Imelda Cheung (Imelda.Cheung@ubc.ca) prior to using the qPCR system.

2.  Turn on the computer and the monitor.

3.  Turn on the C1000 Touch Thermal Cycler (right rear of the instrument).

4.  On the computer, press CTRL-ALT-DELETE to log on.

5.  Type in the following:

  • Login name – LFS-SDW-MC-218:\biorad
  • Psw – maestro1

6.   Once logged in, press “cancel” to activate window prompt, then click “OK”.

7.   Click “Bio-rad CFX Maestro” icon.

8.   On the Startup Wizard Screen, click “User-defined.”

9.   Click “Select Existing” to select a preset method that was saved from your previous runs then click “Open.”

10. If this is the first time you use the instrument, then click “New” to build a new program step by step, following the instructions on screen, and click “save as” and provide a file name.

11. Click “Open Lid” to open lid of the instrument.

12. Load samples in the appropriate location of the instrument.

·     NOTE: Strip tubes must be capped and PCR plate must be sealed.

13. Click “Close Lid” to close lid of the instrument.

14. Click “Run” to initiate the run.

15. When the run is complete, click “save” to ensure data is saved.

16. Export relevant data and graph.


SPECIAL NOTES:

  • Opening and closing of lids MUST BE controlled either by the button on the instrument, or by clicking the “Open Lid” and “Close “Lid” button on the program.
  • NEVER force open or close the lids by force or the lid will become malfunctions.
